The purpose of my thesis was to determine concentrations of certain metals in the roots of interconnected crosses of elderberry. Before the ICP/OES analysis, I had to convert solid samples into a liquid form suitable for analysis. I have done this using the acid digestion technique. For this technique I used HNO3, H2SO4 and H2O2. The samples thus prepared were diluted with 1 % HNO3 and were kept in 50 mL vials.
The samples were ready for the ICP/OES analysis and in the case of magnesium the AAS analysis was used too. Before the analysis of the samples of elderberry roots I performed the calibration, using a multistandard solution VI (Merck). I have determined concentrations of metals in samples from the calibration curve. For magnesium and potassium, the dilution was needed. I have interpreted the concentrations of metals and determined the linearity of the measuring zone.
