In vitro characterization of bioactivities of rhizome and flower extracts of Japanese (Fallopia japonica) and Bohemian (Fallopia x bohemica) knotweed

Abstract
The aim of this research was to compare the antioxidant, anticancer, antidiabetic and antimicrobial activities of Japanese (Fallopia japonica) and Bohemian (Fallopia x bohemica) knotweed rhizome and flower ethanol extracts. Regarding antioxidant activity, rhizome extracts possess the highest antioxidant activity values, while in the assay of cellular antioxidant activity (CAA) with HepG2 cell line, the highest antioxidant activitiy was determined in flowers of F. x bohemica. Since the knotweed extracts are already used in pharmaceutic industry, the possibility of toxicity is very low. The haemolytic assay proved safe the use of our samples towards normal blood cells; moreover, by means of resazurin assay we also checked the possible cytotoxicity on different tissue cell lines, namely three cancer cell lines (HeLa, HepG2 and PaTu) and one non-cancer cell line HEK 293T. We found that the PaTu cell line is the least sensitive towards all knotweed extracts, whereas the HEK 293T cell line is the most sensitive. Flower extracts of both species are less toxic towards cells than the rhizomes ones. The extracts were also able to decrease α-amylase activity in an antidiabetic assay, the rhizome extracts being more effective than the flower ones. Regarding antimicrobial activity, the concentrated ethanol extracts are not able to inhibit bacterial growth; however, they are inhibiting the yeast growth, the rhizome extracts being more effective than the flower ones. We believe that the flowers and rhizomes of knotweeds indicate a new source of antioxidants, which could be used for prevention of diabetes and cancer.

Title:Karakterizacija bioaktivnih učinkov ekstraktov korenik in cvetov japonskega (Fallopia japonica) in češkega (Fallopia x bohemica) dresnika in vitro
Abstract:
Cilj te raziskave je bil primerjava antioksidativne, protirakave, antidiabetične in protimikrobne aktivnosti etanolnih ekstraktov korenik ter cvetov japonskega (Fallopia japonica) in češkega dresnika (Fallopia x bohemica). Rezultati so pokazali, da imajo ekstrakti korenik največjo antioksidativno učinkovitost, medtem ko so se pri testu celične antioksidativne učinkovitosti (CAA) s celično linijo HepG2, ekstrakti cvetov češkega dresnika izkazali za najuspešnejše. Glede na to, da ekstrakte dresnikov že lahko najdemo v obliki kapsul, je verjetnost njihove toksičnosti zelo majhna. Uporabo v farmacevtski industriji jim dovoljuje dejstvo, da niso sposobni razgraditi normalnih krvnih celic, kar smo v naši raziskavi dokazali s testom hemolitične aktivnosti. Nadaljevali smo s poskusom citotoksičnosti z uporabo resazurina. Uporabili smo tri rakave celične linije (HeLa, HepG2 in PaTu) in eno ne-rakavo linijo HEK 293T. PaTu celična linija se je izkazala za namanj občutljivo, za najbolj občutljivo pa linija HEK 293T. Ugotovili smo, da so ekstrakti cvetov japonskega in češkega dresnika manj toksični za celice kot ekstrakti korenik. Vpliv ekstraktov na sladkorno bolezen smo preverili z uporabo encima α-amilaze, kjer so se ekstrakti korenik izkazali za bolj učinkovite od ekstraktov cvetov. Rezultati protimikrobne aktivnosti so pokazali, da koncentrirni etanolni ekstrakti niso zavrli rasti bakterij, zavirali pa so rast kvasovk, pri čemer so bili ekstrakti korenik bolj uspešni od ekstraktov cvetov. Cvetovi in predvsem korenike dresnikov predstavljajo dober in poceni vir antioksidantov, ki bi lahko v prihodnosti preprečevali težave sladkornih in rakavih bolnikov.
