Molekularna in filogenetska opredelitev novih papilomavirusov osamljenih iz novotvorb ustne votline in anogenitalnega predela japonskih makakov

Abstract
Papilomavirusi (PV) so raznolika skupina vrstno-značilnih, majhnih DNA virusov, ki so etiološko povezani z nastankom številnih benignih in malignih neoplastičnih sprememb kožnega ter sluzničnega epitela. Namen naše raziskave je bil opredeliti vsaj en nov genotip PV, ki povzroča okužbe pri japonskih makakih (Macaca fuscata), in ugotoviti ali je bil etiološko povezan z nastankom izhodiščnih novotvorb. V vzorcu benigne novotvorbe anogenitalnega predela japonskega makaka smo pomnožili celotni genom MfuPV2, določili njegovo nukleotidno zaporedje in ga skupaj z genomom MfuPV1, ki je bil osamljen iz vzorca maligne novotvorbe ustne votline japonskega makaka, molekularno in filogenetsko opredelili. Molekularna analiza je pokazala, da oba virusna genoma nosita zapis za beljakovine z ohranjenimi funkcionalnimi domenami in nekodirajoče področje z regulatornimi zaporedji, ki skupaj omogočajo, da lahko PV uspešno zaključita svoj življenjski krog ter okužita nove gostiteljske celice. MfuPV1 se skupaj z genotipi PV, ki so bili osamljeni iz anogenitalne sluznice javanskih in rezus makakov, uvršča v vrsto Alpha-12, medtem ko je genotip MfuPV2 najverjetneje predstavnik nove virusne vrste znotraj rodu Alphapapillomavirus. Na podlagi relativno visokih ocen virusnega bremena MfuPV1 in MfuPV2 sklepamo, da bi lahko bila oba novo-odkrita genotipa PV etiološko povezana z nastankom izhodiščnih novotvorb.

Title:Molecular and phylogenetic characterization of novel papillomaviruses isolated from oral and anogenital neoplasms of Japanese macaques
Abstract:
Papillomaviruses (PV) are a diverse group of species-specific, small DNA viruses, etiologically linked with various benign and malignant neoplasms of cutaneous and mucousal epithelia. The aim of our study was to completely characterize at least one novel PV type, naturally infecting Japanese macaques (Macaca fuscata), and to determine its etiological association with the development of the original neoplastic lesion. The molecular and phylogenetic analyses were performed on complete genome sequences of Macaca fuscata PV type 1 (MfuPV1) and MfuPV2, which have been completely sequenced in a sample of an oral malignant neoplasm and benign anogenital neoplasm of a Japanese macaque, respectively. The molecular analyses revealed that both viral genomes encode characteristic proteins with conserved functional domains and have a non-coding genomic region with regulatory sequences, which enable viruses to complete their life cycles and infect novel host cells. While MfuPV1 together with PVs identified in anogenital mucosa of long-tailed and rhesus macaques clusters within the Alphapapillomavirus species 12, MfuPV2 is most likely a representative of the novel viral species within the genus Alphapapillomavirus. Based on the relatively high viral loads of MfuPV1 and MfuPV2, we believe that both novel PV types could be etiologically linked with original neoplastic lesions.
