Abstract
V raziskavi smo se osredotočili na biološko odstranjevanje fosforja na Centralni čistilni napravi Domžale-Kamnik (CČNDK). Ta je bila v letu 2016 nadgrajena s štirimi sekvenčnimi šaržnimi reaktorji (SBR), ki omogočajo tudi terciarno čiščenje odpadne vode. Namen raziskave je bil ugotoviti ali CČNDK po nadgradnji zadosti novim, strožjim mejnim vrednostim za izpust očiščene vode v odvodnike, ki vstopajo v veljavo z novo Uredbo o odvajanju in čiščenju komunalne odpadne vode (2015). Z ustreznim delovanjem reaktorjev SBR, naj bi CČNDK v prihodnjih letih zagotovila koncentracije skupnega fosforja v iztoku iz čistilne naprave nižje od 1 mg/L. V drugem delu raziskave smo se osredotočili na dokazovanje prisotnosti polifosfat akumulirajočih bakterij (PAO) v aktivnem blatu. V aktivnem blatu iz CČNDK smo prisotnost PAO na različnih stopnjah čiščenja dokazovali preko sledenja funkcionalnega genskega markerja ppk1, ki kodira encim polifosfat kinazo 1, ključno za sintezo polifosfata iz molekule ATP. Poleg tega smo z oligonukleotidnimi začetniki, ki nalegajo na specifično regijo gena za 16S rDNK posebej sledili tudi domnevno ključnega predstavnika PAO - še neizolirano bakterijsko vrsto Candidatus Accumulibacter phosphatis (CAP). S kemijskimi analizami odpadne vode smo ugotovili, da je CČNDK z nadgradnjo v letu 2016 zadostila novim, strožjim mejnim vrednostim. Ugotovili smo, da je bila v zagonski fazi mikrobiološka učinkovitost čiščenja fosforja iz odpadne vode 35 %. Z molekularnimi metodami smo dokazali prisotnost PAO v aktivnem blatu odvzem na različnih merilnih mestih.

Title:Assesment of microbiological phosphorus removal efficiency from wastewater in early working stage of SBR bioreactor
Abstract:
In this research we focused on biological phosphorus removal in Wastewater treatment plant Domzale-Kamnik (WWTPDK). In 2016 WWTPDK was upgraded with four sequence batch reactors (SBR), which enable tertiary treatment of wastewater. The aim of our research was to find out, if the upgrade of WWTPDK reduces the amounts of phosphorous in wastewaters below the values established in a new regulatory limits. With appropriate functioning, SBR reactors should reduce the total phosphorus concentrations in WWTPDK effluent below 1 mg/L. In second part of the research, we tried to detect phosphate accumulating organisms (PAO) in the active sludge. PAO presence in active sludge from different parts of WWTPDK was confirmed via the presence of functional genetic marker ppk1. The latter encodes the enzyme crucial for synthesis of polyphosphate from ATP - polyphosphate kinase 1. Specific 16S rDNA primers were also used to identify important yet unisolated representative of PAO Candidatus Accumulibacter phosphatis (CAP). Chemical analysis of wastewater proved that the upgraded WWTPDK can satisfactory reduce the amounts of phosphorous below the limiting values established in a new regulation. Microbiological phosphorus removal efficiency in wastewater was 35 %. We also detected PAO in the active sludge from different sampling points.
