Europe is the most important producer and exporter of vine seedlings (Vitis vinifera L.) and wine. This important economic section is endangered by many different diseases, fungi diseases and grapevine yellows (GY) diseases. Responsible for GY diseases are phytoplasmas, phytopathogenic bacteria from class Mollicutes. In Europe there are two major phytoplasma groups that cause GY. 'Candidatus Phytoplasma solani' which is associated with yellows disease ('Bois noir', BN). And second yellows disease (Flavescence dorée, FD) which is associated with quarantine phytoplasma FDp. Using bioinformatic tools, we chose from effector catalogue, which was prepared by dr. Günter Brader scientific group from Austrian institute for technology, six potencial effectors. We inserted sequences of chosen effectors in to binary vectors and agroinfiltrated them in to tobacco leaves.. Using confocal microscopy, we showed that all six effectors localized in nucleous and cytoplasma of transformed cells. We tried to introduce a grapevine Agrobacterium-mediated transient transformation protocol to our lab.
