Mesenchymal stem cells MSC are cells, which help us in treating various injuries and deseases in clinic. Because of their immunosupressiveness, they are important factor in GvHD treatment, which may occur after allogenic tissue transplantation. We isolated immune cells PBMC from two different donors. Isolation was made from the buffy coat, a suspension of leukocytes, granulocytes and still some erytrocytes. We created a mixed lymphocyte reaction, where we aded five different concentration of 30 Gy irradiated MSC. The thymidine reagent, which was used, was incorporated into the immune cells. From an amount which was mesured, we concluded proliferation of PBMC. With proliferation, we detected an inhibitory effect of MSC to immune cells. Optimal MSC number was 10⁵. In the case of higher quantity, there would be a contact inhibition, which would cause cells to die. Proliferation with increased MSC quantity, was dropping disproportinately.
